ABSTRACT

El extracto de cloroformo (CE) y las fracciones obtenidas de las raíces de Aldama arenaria se evaluaron para determinar su actividad antiproliferativa in vitro contra 10 líneas celulares tumorales humanas [leucemia (K-562), mama (MCF-7), ovario que expresa un fenotipo resistente a múltiples fármacos (NCI/ADR-RES), melanoma (UACC-62), pulmón (NCI-H460), próstata (PC-3), colon (HT29), ovario (OVCAR-3), glioma (U251) y riñón (786-0)]. CE presentó actividad antiproliferativa débil a moderada (log GI50 medio 1.07), mientras que las fracciones 3 y 4, enriquecidas con diterpenos de tipo pimarane [ent-pimara-8 (14), ácido 15-dien-19-oico y ent-8(14),15-pimaradien-3ß-ol], presentaron actividad moderada a potente para la mayoría de las líneas celulares, con un log GI50 medio de 0.62 y 0.59, respectivamente. Los resultados mostraron una acción antiproliferativa in vitro prometedora de las muestras obtenidas de A. arenaria, con los mejores resultados para NCI/ADR-RES, HT29 y OVCAR-3, y valores de TGI que van desde 5.95 a 28.71 µg.mL-1, demostrando que los compuestos de esta clase pueden ser prototipos potenciales para el descubrimiento de nuevos agentes terapéuticos.


Chloroform extract (CE) and fractions obtained from Aldama arenaria roots were evaluated for their in vitro antiproliferative activity against 10 human tumor cell lines [leukemia (K-562), breast (MCF-7), ovary expressing a multidrug-resistant phenotype (NCI/ADR-RES), melanoma (UACC-62), lung (NCI-H460), prostate (PC-3), colon (HT29), ovary (OVCAR-3), glioma (U251), and kidney (786-0)]. CE presented weak to moderate antiproliferative activity (mean log GI50 1.07), whereas fractions 3 and 4, enriched with pimarane-type diterpenes [ent-pimara-8(14),15-dien-19-oic acid and ent-8(14),15-pimaradien-3ß-ol], presented moderate to potent activity for most cell lines, with mean log GI50 of 0.62 and 0.59, respectively. The results showed promising in vitro antiproliferative action of the samples obtained from A. arenaria, with the best results for NCI/ADR-RES, HT29, and OVCAR-3, and TGI values ranging from 5.95 to 28.71 µg.mL-1, demonstrating that compounds of this class may be potential prototypes for the discovery of new therapeutic agents.

ABSTRACT

This study aimed to present information about the sarsaparilla sold in establishments in the state of São Paulo, Brazil, assess the genetic diversity of Smilax brasiliensis Spreng., Smilacaceae, and examine the growing conditions and productivity of five species of Smilax. The amount of sarsaparilla sold per month at most pharmacies was 0.4 kg on average. Herbal stores and markets sold averages of 9 kg and 8 kg per month, respectively. The weight of the underground biomass of S. fluminensis (188.3 g) is significantly higher than those of other species (28.3-79.6 g). The study demonstrated that high genetic diversity among the Smilax brasiliensis plants belonging to the CPQBA germplasm bank, which was confirmed by the results of the genotyping study that used a SSR marker on S. brasiliensis. The high consumption of sarsaparilla and the low yield of young plants cultivated from seeds with high genetic variability reinforce the need for further studies on the production of Smilax species.

ABSTRACT

This study aimed to describe the morphoanatomy of the aerial vegetative organs of seven Smilax species, used in Brazilian folk medicine. Samples of leaves and stems were fixed with FAA 50, embedded in historesin, sectioned on a rotary microtome, stained and mounted in synthetic resin. Cuticle ornamentation was analyzed with standard scanning electron microscopy. In the frontal view, the walls of the adaxial epidermis are straight in S. brasiliensis, S. cissoides, S. goyazana and sinuous in the other species. The walls of the epidermis on the abaxial surface are straight in S. brasiliensis, S. goyazana, S. rufescens, sinuous in S. campestris, S. fluminensis, S. oblongifolia, and wavy in S. cissoides. The stomata are paracytic in S. brasiliensis, S. goyazana, S. oblongifolia, and S. rufescens, anomocytic in S. cissoides, S. campestris; anisocytic and paracytic in S. fluminensis. The midrib has three vascular bundles that are individually wrapped by lignified cells in S. brasiliensis, S. cissoides, and S. fluminensis. In the other, the three vascular bundles are surrounded by a single lignified sheath. In the stems the vascular cylinder is surrounded by a sclerenchymatous ring with the exception of Smilax fluminensis, which has a starch sheath and internal layers of thin-walled cells.

ABSTRACT

Thiamethoxam is a systemic insecticide from the neonicotinoid group, nitroguanidin family which affects the nicotinic receptor acetyl choline in the insect membrane, wounding the nervous system and causing the death of the insect. It was used with success in the control of initial pests of several crops. It was considered that thiamethoxam has a bioactivator effect, because in the absence of insects promoted increase in vigor, development and productivity of crops. This work was carried out to verify if thiamethoxam causes histological changes in sugarcane roots. In this work, it was used optical microscopy, images arrest, tissue biometrics and statistical analysis, in young roots of sugarcane RB 83 5486 after the treatments with different thiamethoxam concentrations. It was determined changes in histological structure of tissues 7, 14, 21 and 28 days after the treatments, establishing its effects on root plant anatomy. It was verified that thiamethoxam increased root cortex width, increasing the vascular cylinder and the metaxylem vessel elements number in the vascular tissue until 21 days after application.


O tiametoxam é um inseticida sistêmico do grupo neonicotinoide, da família nitroguanidina que atua no receptor nicotínico acetilcolina da membrana de insetos, lesando o sistema nervoso e levando-os à morte. Tem sido utilizado com sucesso no controle de pragas iniciais de diversas culturas. Considerou-se que o produto possui um efeito bioativador, uma vez que, mesmo na ausência dos insetos-praga, tem promovido aumento em vigor, desenvolvimento e produtividade de cultivos. No presente trabalho, foi analisada a anatomia de raízes jovens de cana-de-açúcar RB 83 5486 após aplicações de tiametoxam nas concentrações de 0, 400 e 800g ha-1. As análises foram conduzidas aos 7, 14, 21 e 28 dias após as aplicações (DAA), no sentido de esclarecer seus efeitos na anatomia radicular da planta. Verificou-se que, até os 21DAA de desenvolvimento da cana-de-açúcar, thiametoxam amplia a espessura do córtex da raiz e aumenta o diâmetro do cilindro vascular, bem como o número de elementos de vaso do metaxilema, melhorando a absorção de água e sais minerais.

This study aimed to analyze the cuticle thickness and pattern of epicuticular wax deposition in 'Gala' and 'Galaxy' apple cultivars (Malus domestica Borkh,) from three Brazilian producing areas: Vacaria (RS), Fraiburgo (SC) and São Joaquim (SC) with altitudes of 971, 1,048 and 1,353m, respectively. Harvested fruit were kept under two storage conditions: regular atmosphere (RA) (0 ºC and 90% RH) and controlled atmosphere (CA) (1.5% O2, 2.5% CO2, 0ºC and 90% RH). Cuticle thickness measurements were made using LM and the deposition pattern of epicuticular wax observed with a SEM. Altitude among the apple producing areas was not a factor in deposition pattern of waxes between the cultivars but at higher altitudes, the cuticle was thicker in both the cultivars. In the freshly-harvested fruits, waxes deposition in the form of platelets and the mechanism of “tear and repair” were observed. Severity of microcracks in the cuticle was more evident on the fruits from CA.

Smilax L. in Brazil is represented by 32 taxa and it is a taxonomically difficult genus because the plants are dioecious and show wide phenotypic variation. The analysis and use of leaf anatomy characters is recognized as a frequently successful taxonomic method to distinguish between individual taxon, when floral material is absent or minute differences in flowers and foliage exist such as in Smilax. The aim of this study was to characterize the anatomical features of the aerial organs in Smilax syphilitica collected from the Atlantic Rainforest, in Santa Teresa-ES and the Smilax aff. syphilitica from the Amazon Rainforest, in Manaus, Brazil. For this, a total of three samples of Smilax were collected per site. Sample leaves and stems were fixed with FAA 50, embedded in historesin, sectioned on a rotary microtome, stained and mounted in synthetic resin. Additionally, histochemical tests were performed and cuticle ornamentation was analyzed with standard scanning electron microscopy. S. syphilitica and S. aff. syphilitica differed in cuticle ornamentation, epidermal cell arrangement and wall thickness, stomata type and orientation, calcium oxalate crystal type, and position of stem thorns. Leaf blades of S. syphilitica from the Amazon Rainforest have a network of rounded ridges on both sides, while in S. aff. syphilitica, these ridges are parallel and the spaces between them are filled with numerous membranous platelets. Viewed from the front, the epidermal cells of S. syphilitica have sinuous walls (even more pronounced in samples from the Amazon); while in S. aff. syphilitica, these cells are also sinuous but elongated in the cross-section of the blade and arranged in parallel. Stomata of S. syphilitica are paracytic, whereas in S. aff. syphilitica, are both paracytic and anisocytic, and their polar axes are directed towards the mid-vein. Calcium oxalate crystals in S. syphilitica are prisms, whereas in S. aff. syphilitica, crystal sand. Thorns occur in nodes and internodes in S. syphilitica but only in internodes in S. aff. syphilitica. These features have proven to be of diagnostic value and may support a separation into two species, but future studies are needed to confirm that S. aff. syphilitica is indeed a new taxon.


Smilax L. en Brasil está representado por 32 táxones y es un género difícil en lo que respecta a su taxonomía porque las plantas son diocas y presentan mucha variación fenotípica. El análisis y uso de los caracteres anatómicos de la hoja es reconocido frecuentemente como un exitoso método taxonómico para distinguir entre los táxones individuales cuando las flores están ausentes o hay diminutas diferencias. El objetivo de este estudio fue caracterizar los rasgos anatómicos de los órganos aéreos en Smilax syphilitica recolectados en el Bosque Atlántico en Santa Teresa- ES y de Smilax aff. syphilitica del Amazonas, Manaus, Brasil. Un total de tres muestras fueron recolectadas por sitio. Las muestras fueron fijadas en FAA 50, sumergidas en historesina, seccionadas en un micrótomo rotatorio, teñidas y montadas en resina sintética. Se hicieron pruebas histoquímicas y la ornamentación de la cutícula fue analizada con microscopía electrónica de barrido estándar. S. syphilitica y difirieron en la ornamentación de la cutícula, la disposición de células epidérmicas y el grosor de la pared, tipo y orientación de estomas, tipo de cristal de oxalato de calcio y la posición de las espinas del tallo. Estas características han probado ser de valor diagnóstico y pueden apoyar la separación de dos especies, pero se necesitan futuros estudios para confirmar que S. S. aff. syphiliticaaff. syphilitica es un nuevo taxon.

Species of Smilax, also known as greenbrier, are widely distributed in Brazil and their commercial trades are carried out by the extractivism of native species. We the aim to provide information about the germination and development of seedlings in four Smilax species, different experiments were developed under controlled conditions. We evaluated two germination treatments: temperature (30ºC and 20-30ºC) and light (presence/ absence), and for few cases the tetrazolium treatment was applied. A different treatment response was observed among the studied species. Light had a significant influence in S. brasiliensis, with the highest germination rates at 20-30ºC in dark conditions. S. campestris showed significant differences among temperature treatments, but not to light; while S. cissoides showed high germination rates (66-78%), independently of treatment. However, S. polyantha had low germination rates (19-24%). After one year, the expanded leaves showed different characteristics among the studied species. Leaves of S. brasiliensis were ovate, coriaceous, three main veins and prickle-like structures only on the midrib on abaxial face. S. campestris leaves were oblong, coriaceous and prickle-like structures were located at the leaf midrib and margin. S. cissoides had ovate-elliptic, membranaceous leaves, with three main veins with prickle-like structures on the abaxial face. S. polyantha leaves showed ovateelliptic, coriaceous leaves, with three main veins, translucent secondary veins and no prickle-like structures. A seedling identification key was elaborated based on morphological characteristics.

Smilax L. species, popularly known in Brazil as salsaparrilha, have been used in folk medicine as tonic against rheumatism and as anti-syphilitic since the sixteenth century; however, until today, their roots have been explored in an extractive way. The aim of this study was to analyze the propagation of Smilax fluminensis aiming to meet the demand and to propose procedures for its sustainable management. It was analyzed the percentage of seed germination, rooting of aerial and subterranean stem cuttings and plant resprouting capacity after removal of the shoots. The monitoring lasted one year. Seeds were subjected to temperatures: 25°C, 30°C, 20-30°C and 20-35°C under a daily photoperiod of eight hours and in the absence of light. For rooting the stem cuttings, aerial and subterranean stems about 20cm long with two nodal regions were subjected to treatment with distilled water (control) and indolbutyric acid (IBA) at 100ppm. The best germination percentages obtained for S. fluminensis were 80 percent at 20-30°C under light and 85 percent at 30°C in the dark. Only subterranean stem cuttings showed significant difference concerning fresh and dry matter of roots with higher values in treatments with hormone compared to the control.


As espécies do gênero Smilax L., conhecidas popularmente como salsaparrilha, são empregadas na medicina popular como fortificante contra o reumatismo e antissifilítico desde o século XVI, porém até hoje as suas raízes são exploradas de maneira extrativista. O objetivo deste estudo foi analisar a propagação de Smilax fluminensis visando a auxiliar no atendimento da demanda e apontar propostas para o seu manejo sustentável. Foram analisados a porcentagem de germinação de sementes, o enraizamento de estacas de ramos aéreos e subterrâneos e o acompanhamento ao longo de um ano da capacidade de rebrotamento das plantas após a retirada dos ramos aéreos. As sementes foram submetidas às temperaturas: 25, 30, 20-30 e 20-35°C, sob fotoperíodo diário de oito horas e na ausência de luz. Para o enraizamento de estacas, foram utilizados ramos aéreos e subterrâneos com aproximadamente 20cm com duas regiões nodais submetidas ao tratamento com água destilada (controle) e com ácido indolbutírico (AIB) a 100ppm. As melhores porcentagens de germinação obtidas para S. fluminensis, 80 por cento em 20-30°C no claro e 85 por cento a 30°C no escuro. Apenas as estacas de ramos subterrâneos enraizaram, houve diferença significativa em relação à massa fresca e seca de raízes com valores maiores nos tratamentos com hormônio em relação ao controle.

In the Brazilian Cerrado (neotropical savanna), the development of bud-bearing underground systems as adaptive structures to fire and dry periods can comprise an important source of buds for this ecosystem, as already demonstrated in the Brazilian Campos grasslands and North American prairies. Asteraceae species from both woody and herbaceous strata have subterranean organs that accumulate carbohydrates, reinforcing the adaptive strategy of these plants to different environmental conditions. This study aims to analyse the morpho-anatomy of underground systems of six species of Asteraceae (Mikania cordifolia L.f. Willd., Mikania sessilifolia DC, Trixis nobilis (Vell.) Katinas, Pterocaulon alopecuroides (Lam.) DC., Vernonia elegans Gardner and Vernonia megapotamica Spreng.), to describe these structures and to verify the occurrence and origin of shoot buds, and to analyse the presence of reserve substances. Individuals sampled in Cerrado areas in São Paulo State showed thick underground bud-bearing organs, with adventitious or lateral roots and presence of fructans. Xylopodium was found in all studied species, except for Trixis nobilis, which had stem tuber. The presence of fructans as reserve, and the capacity of structures in the formation of buds indicate the potential of herbaceous species of Asteraceae in forming a viable bud bank for vegetation regeneration in the Brazilian Cerrado.


No Cerrado brasileiro (savana neotropical), o desenvolvimento de sistemas subterrâneos que produzem gemas, como estruturas adaptativas contra o fogo e períodos de seca, pode compreender um importante suprimento de gemas para esse ecossistema, como já demonstrado nos campos brasileiros e nas pradarias norte-americanas. Espécies de Asteraceae tanto do estrato lenhoso, quanto do herbáceo têm órgãos que acumulam carboidratos, reforçando a estratégia adaptativa dessas plantas a diferentes condições ambientais. Este estudo tem o objetivo de analisar a morfo-anatomia de sistemas subterrâneos de seis espécies de Asteraceae (Mikania cordifolia L.f. Willd., Mikania sessilifolia DC, Trixis nobilis (Vell.) Katinas, Pterocaulon alopecuroides (Lam.) DC., Vernonia elegans Gardner e Vernonia megapotamica Spreng.), para descrever essas estruturas e verificar a ocorrência e origem de gemas caulinares, e analisar a presença de substâncias de reserva. Indivíduos amostrados em áreas de Cerrado no Estado de São Paulo apresentaram órgãos subterrâneos espessados produtores de gemas, com raízes adventícias ou laterais e presença de frutanos. Em todas as espécies estudadas foi constatada a presença de xilopódio, com exceção de Trixis nobilis, a qual apresentou caule tuberoso. A presença de frutanos como reserva e a capacidade de estruturas na formação de gemas indicam o potencial de espécies herbáceas de Asteraceae em formar um banco de gemas viável para regeneração da vegetação no Cerrado brasileiro.

The leprosis disease shows a viral etiology and the citrus leprosis virus is considered its etiologic agent. The disease may show two types of cytopatologic symptom caused by two virus: nuclear (CiLV-N) and cytoplasmic (CiLV-C) types. The aim of this study was to compare the morpho-anatomical differences in the lesions caused by leprosis virus-cytoplasmic and nuclear types in Citrus sinensis (L.) Osbeck 'Pêra'. Leaf and fruit lesions were collected in Piracicaba/São Paulo (cytoplasmic type) and Monte Alegre do Sul/São Paulo and Amparo/São Paulo (nuclear type). The lesions were photographed and then fixed in Karnovsky solution, dehydrated in a graded ethylic series, embedded in hydroxy-ethyl methacrylate resin (Leica Historesin), sectioned (5 μm thick), stained and mounted in synthetic resin. The digital images were acquired in a microscope with digital video camera. Leaf and fruit lesions caused by the two viruses were morphologically distinct. Only the lesion caused by CiLV-N virus presented three well-defined regions. In both lesions there was the accumulation of lipidic substances in necrotic areas that were surrounded by cells with amorphous or droplets protein. Only leaf and fruit lesions caused by CiLV-N virus exhibited traumatic gum ducts in the vascular bundles.


A doença leprose dos citros tem etiologia viral sendo o citrus leprosis virus seu agente etiológico. Demonstrou-se que há dois vírus distintos que causam sintomas de leprose em ci-tros: citoplasmático (CiLV-C) e o nuclear (CiLV-N). O objetivo desse estudo foi comparar as diferenças morfo-anatômicas nas lesões causadas por CiLV-C e por CiLV-N em laranjeira doce (Citrus sinensis (L.) Osbeck) 'Pêra'. As lesões foliares e dos frutos foram coletadas em Piracicaba/SP (tipo citoplas-mático) e em Monte Alegre do Sul/SP e Amparo/SP (tipo nuclear). As lesões foram fotografadas e em seguida fixadas em solução Karnovsky, desidratadas em série etílica, incluídas em historesina e secionadas em micrótomo rotativo. As lâminas foram coradas, analisadas e fotografadas. As lesões foliares e do fruto causadas pelos dois vírus eram morfologicamente distintas. Somente a lesão causada por CiLV-N apresentou três regiões bem definidas. Em ambas as lesões ocorreu acúmulo de substâncias lipídicas nas áreas necrosadas que se achavam envoltas por células com conteúdo protéico amorfo ou em gotas. Somente as lesões da folha e do fruto causadas pelo CiLV-N exibiram ductos traumáticos gomosos nos feixes vasculares.

Previous studies pointed out that species richness and high density values within the Leguminosae in Brazilian forest fragments affected by fire could be due, at least partially, to the high incidence of root sprouting in this family. However, there are few studies of the factors that induce root sprouting in woody plants after disturbance. We investigated the bud formation on root cuttings, and considered a man-made disturbance that isolates the root from the shoot apical dominance of three Leguminosae (Bauhinia forficata Link., Centrolobium tomentosum Guill. ex Benth, and Inga laurina (Sw.) Willd) and one Rutaceae (Esenbeckia febrifuga (St. Hil.) Juss. ex Mart.). All these species resprout frequently after fire. We also attempted to induce bud formation on root systems by removing the main trunk, girdling or sectioning the shallow lateral roots from forest tree species Esenbeckia febrifuga and Hymenaea courbaril L. We identified the origin of shoot primordia and their early development by fixing the samples in Karnovsky solution, dehydrating in ethyl alcohol series and embedding in plastic resin. Serial sections were cut on a rotary microtome and stained with toluidine blue O. Permanent slides were mounted in synthetic resin. We observed different modes of bud origin on root cuttings: close to the vascular cambium (C. tomentosum), from the callus (B. forficata and E. febrifuga) and from the phloematic parenchyma proliferation (I. laurina). Fragments of B. forficata root bark were also capable of forming reparative buds from healing phellogen formed in callus in the bark’s inner side. In the attempt of bud induction on root systems, Hymenaea courbaril did not respond to any of the induction tests, probably because of plant age. However, Esenbeckia febrifuga roots formed suckers when the main trunk was removed or their roots were sectioned and isolated from the original plant. We experimentally demonstrated the ability of four tree species to resprout from roots after disturbance. Estudios anteriores señalaron que la alta riqueza de especies de Leguminosae en fragmentos de bosques afectadas por el fuego podría deberse, al menos parcialmente, a la alta incidencia de brotes de raíz en esta familia. Sin embargo, existen pocos estudios. Investigamos la formación de brotes en tres Leguminosae (Bauhinia forficata Link., Centrolobium tomentosum Guill. ex Benth, e Inga laurina (Sw.) Willd) y una Rutaceae (Esenbeckia febrifuga (St. Hil.) Juss. ex Mart.). Todas estas especies suelen rebrotar después de los incendios. También hicimos cortes experimentales en árboles forestales: Esenbeckia febrifuga e Hymenaea courbaril L. Observamos tres orígenes: cerca del cambium vascular (C. tomentosum), desde el callo (B. forficata y E. febrifuga) y de parénquima (I. laurina). Hymenaea courbaril no respondió a ninguna de las pruebas de inducción, probablemente debido a la edad de la planta. Sin embargo, las raíces de Esenbeckia febrifuga forman retoños tras retirar el tronco o separar sus raíces. Hemos demostrado experimentalmente la capacidad de las cuatro especies de árbol de rebrotar a partir de sus raíces tras una perturbación grave.

ABSTRACT

This work dealt with the anatomy of the underground system in Vernonia grandiflora Less. and V. brevifolia Less. (Vernonieae; Asteraceae), two perennial geophytes, to elucidate their ability to sprout in the Brazilian Cerrado conditions. V. grandiflora, a subshrubby species, possessed a thickened underground system constituted by a xylopodium and many tuberous roots. The xylopodium had stem and root structure and its buds were axillary or originated from the cortical parenchyma proliferation. The tuberous roots produced by this organ were adventitious and accumulated inulin-type fructans mainly in the cortical parenchyma. The thickened underground system of V. brevifolia, an herbaceous species, was a tuberous primary root whose buds originated from the proliferated pericycle. The occurrence of these bud-forming underground systems, which stored reserve compounds, enabled these plants to survive throughout unfavourable environmental conditions in the Cerrado, such as dry season and frequent fires in the winter.


Este trabalho teve como objetivo estudar a anatomia dos sistemas subterrâneos de Vernonia grandiflora Less. e V. brevifolia Less. (Vernonieae; Asteraceae), duas geófitas perenes, a fim de esclarecer sua capacidade para brotar em condições de Cerrado. O sistema subterrâneo espessado de V. grandiflora, uma espécie subarbustiva, é constituído pelo xilopódio e por várias raízes tuberosas. O xilopódio possui estrutura mista (radicular e caulinar) e suas gemas são de origem axilar ou se originam a partir da proliferação do parênquima cortical. As raízes tuberosas produzidas por este órgão são adventícias e acumulam frutanos do tipo inulina, principalmente no parênquima cortical. Em V. brevifolia, uma espécie herbácea, o sistema subterrâneo espessado é constituído pela raiz primária cujas gemas são originadas a partir do periciclo proliferado. A ocorrência destes sistemas subterrâneos gemíferos, que armazenam compostos de reserva, permite que estas plantas sobrevivam às condições desfavoráveis do Cerrado, tais como a estação seca e os freqüentes incêndios durante o inverno.

ABSTRACT

Different concentrations of 6-benzyladenine (6-BA) were used in vitro to induce buds in three types of explants: leaf discs, root segments and the seedling itself obtained from in vitro seed germination of Passiflora cincinnata Mast. The 0.5 mg.L -1 6-BA concentration was most suitable for all the three explants, however, bud formation time and means (direct/indirect) were different for each type of explant. In seedlings used as explants, it had shoot regeneration from the primary root. Histological events leading to bud formation via meristemoids were described. The origin was indirect in leaf discs, with callus formation in subepidermal cells of the chlorophyll parenchyma layers. In root segments and in seedling roots, the buds were formed directly in the pericycle (in roots with some secondary structure) and in the vascular cambium (at the initial phase of secondary structure). Also, indirect buds originated from meristemoids which were formed around the callus.


Diferentes concentrações de 6-benziladenida (6-BA) foram usadas para a indução de brotos in vitro em três tipos de explantes: discos foliares, segmentos de raiz e plântula obtidos da germinação de sementes in vitro de Passiflora cincinnata Mast. A concentração de 0,5 mg.L-1 foi a mais adequada para os três explantes, porém, o tempo e a via (direta/indireta) de formação da gema diferiu para cada tipo de explante. Nas plântulas utilizadas como explante houve a formação de gemas a partir da raiz primária. Os eventos histológicos que levaram a formação de um novo órgão, via meristemóides (centros meristemáticos) foram descritos. Nos discos foliares a origem foi indireta, com formação de calo a partir das células subepidérmicas das camadas de parênquima clorofiliano. Nos segmentos de raiz e nas raízes das plântulas, a organogênese direta apresentou duas origens, a partir do periciclo, nas raízes com início da estrutura secundária, e a partir do câmbio vascular, nas raízes com estrutura secundária já estabelecida. Também nos segmentos de raiz observou-se a via indireta cujo calo resultou da proliferação do periciclo.

ABSTRACT

Este trabalho teve por objetivo descrever e comparar a morfo-anatomia das folhas adultas de Mikania glomerata Spreng., Porophyllum ruderale Cass. e Vernonia condensata Baker (Asteraceae), que possuem diferentes hábitos, enfatizando suas estruturas secretoras. Secções longitudinais e transversais dos limbos foliares foram analisadas nas regiões do ápice, da base e do terço médio na altura da nervura central, do limbo foliar e da margem. M. glomerata apresentou tricomas glandulares unisseriados e ductos secretores; P. ruderale tinha hidatódios e cavidades secretoras; e V. condensata apresentou idioblastos e tricomas glandulares uni- ou bisseriados.

ABSTRACT

Plant regeneration was achieved from cells of callus induced from hypocotyl segments of Bauhinia forficata on half strength Murashige and Skoog culture medium supplemented with several concentrations of BAP. Within 40 days of culture shoot buds formation was observed on callus surface. Calli were then transferred to a same composition culture medium without plant growth regulator in order to induce shoot elongation. Histological studies indicated that in vitro plant regeneration in B. forficata occurred through indirect organogenesis. Meristemoids consisting of small cells with dense cytoplasm and prominent nuclei were randomly distributed throughout the callus surface indicating early stages of shoot bud differentiation. Shoots developed de novo from superficial layers of cells and the pattern of shoot origin and development were very similar to those previously described for other leguminous species.


A regeneração de plantas de Bauhinia forficata a partir de calos induzidos em segmentos de hipocótilo foi obtida em meio de cultura contendo metade da formulação de sais de Murashige & Skoog, suplementado com várias concentrações de BAP. Após 40 dias de cultura, ocorreu a formação de gemas adventícias na superfície do calo. Os calos que apresentavam tais formações, foram então transferidos para o mesmo meio de cultura mas sem a adição de fitorreguladores, visando o alongamento das gemas. Estudos histológicos indicaram que a regeneração in vitro de plantas de B. forficata ocorre pela via organogenética. Meristemóides caracterizados por células pequenas, de citoplasma denso e núcleo e nucléolo proeminentes se encontravam distribuídos pela superfície do calo, caracterizando os estágios iniciais da diferenciação de gemas caulinares. Partes aéreas se desenvolveram de novo a partir de camadas superficiais de células e seu padrão de origem e desenvolvimento foi muito similar aos previamente descritos na literatura para outras espécies de leguminosas.
